Análisis del efecto del tratamiento magnético sobre la concentración de proteínas totales en semillas de tomate (Solanum lycopersicum L.)

spa:La estimulación de semillas y de plántulas a través de campos magnéticos, electromagnéticos, infrarrojo, rayos gama, rayos láser y rayos U.V. disminuyen los riesgos ambientales y sanitarios, evitando el uso de agroquímicos. La exposición de semillas a campos magnéticos es valiosa, dado que en investigaciones anteriores se ha observado que el tiempo de germinación de las semillas se reduce, la longitud de la radícula, el área foliar y la actividad enzimática aumentan. Sin embargo, aún no se conoce claramente como la estimulación magnética genera alteraciones en los procesos bioquímicos de la semilla, por lo tanto, es necesario investigaciones de esta índole para comprender las respuestas fisiológicas, las modificaciones en la morfología y algunos procesos celulares. En este estudio se efectuaron ocho tratamientos magnéticos en semillas de tomate con un conjunto de imanes toroidales con densidad de flujo magnético nominal de. El análisis de los tratamientos se realizó empleando una optimización bayesiana, teniendo como variables el tiempo de exposición (Texp),temperatura de incubación y la cantidad de agua, siendo los intervalos 0.0 a 900 s, 20.0 a 35.0 ºC y 3.0 a 15.0 mL respectivamente. El efecto magnético sobre la germinación como variable biológica se evaluó midiendo el Tiempo Medio de Germinación (TMG) y como variable química determinado la concentración de proteínas totales. Las semillas germinadas se someterán a dos procesos de extracción de proteínas: agitación orbital y ultrasonido empleando como solvente Tris-HCl o Tris-Base y posteriormente la cuantificación se realizará por medio del método espectrofotométrico UV-visible de Bradford. La mejor metodología de extracción para la obtención de proteínas totales fue empleando ultrasonido como medito físico, tris-base como solvente y un tiempo de extracción de 50 min. Las mejores dosis de estimulación magnética fueron obtenidas mediante el diseño de optimización bayesiana presentando TMG inferiores a las . Al comparar la concentración de proteínas totales entre los tratamientos con los mejores TMG y el control, se determinó que seis tratamientos tuvieron concentraciones por encima del control, de los cuales cuatro presentaron diferencias estadísticamente significativas entre ellos. eng:The stimulation of seeds and seedlings through magnetic, electromagnetic, infrared, gamma, laser and U.V. fields reduce environmental and sanitary risks, avoiding the use of agrochemicals. The exposure of seeds to magnetic fields is valuable, since in previous research it has been observed that seed germination time is reduced, radicle length, leaf area and enzymatic activity are increased. Positive results have also been found in response to the activation of biological, biophysical and biochemical mechanisms of action. However, it is still not clearly known how magnetic stimulation generates alterations in the biochemical processes of the seed, therefore, research of this nature is necessary to understand the physiological responses, modifications in morphology and some cellular processes. In this study, eight magnetic treatments were performed on tomato seeds with a set of toroidal magnets with a nominal magnetic flux density of 100.00 mT. The analysis of the treatments was performed using a Bayesian optimization, having as variables the exposure time (Texp), incubation temperature and amount of water, the intervals being 0.0 to 900 s, 20.0 to 35.0 ºC and 3.0 to 15.0 mL, respectively. The magnetic effect on germination as a biological variable was evaluated by measuring the Mean Germination Time (MTG) and as a chemical variable, the concentration of total proteins was determined. The germinated seeds were subjected to two protein extraction processes: orbital shaking and ultrasound using Tris-HCl or Tris-Base as solvent, and then quantification was performed by means of the Bradford UV-visible spectrophotometric method. The best extraction methodology to obtain total proteins was using ultrasound as physical medium, tris-base as solvent and an extraction time of 50 min. The best doses of magnetic stimulation were obtained by Bayesian optimization design presenting TMG lower than 65.00 hours. When comparing the total protein concentration between the treatments with the best TMG and the control, it was determined that six treatments had concentrations above the control, of which four showed statistically significant differences between them.
